Cyclins contain two domains of a similar all- alpha fold, the first located at the N- terminus and the second at the C- terminus. Cyclin contain a tertiary structure of two compact domains of 5 alpha helices. The first of which is conserved cyclin box, outside of which cyclin are divergent.

Regulation of CDKs by phosphorylation cyclin binding to the CDK contributes to the kinase activation by inducing a conformational change wherein a C- terminal domain of the CDK referred to as the T- loop is directed out of the substrate binding cleft.
