Translation begin when the small subunit of ribosome recognise the 5' cap of the mRNA in case of eukaryotes and scan the mRNA until it encounter the start codon. Once it recognised the start codon the initiator tRNA bind to the p site and during elongation the incoming tRNA bind to the A site .
Peptide bond formation occur between the amino acid bound to tRNA in the p site to the amino acid bound to tRNA in the A site.
Start codon codes for methionine as a result first amino acid in the polypeptide chain is methionine.
Origin of replication and primer are part of replication not translation. +1 start site is a part of transcription and not translation. 5' cap is a part of translation but not involve in initiating the translation.
