Co-culture experiments were carried out on PDL-coated glass coverslips (PDL being the best culture substrate for mucosa-derived OECs out of those tested in this study – PDL, laminin and TCP) in a 24-well plate. NG108-15 neurons were either grown alone, on a monolayer of mucosa-derived cells or on a monolayer of Schwann cells .

First 104 mucosa-derived cells or Schwann cells were seeded onto the coated coverslip and allowed to adhere overnight, before 103 NG108-15 neurons were seeded on the monolayer. The co-culture was fixed 5 days later with 4% PFA at 4 °C overnight. Media was changed every second day: DMEM/F12 + 1% P/S + 10% FBS.
