Cells (suspended in a buffer)are made to break with some physical attrition and centrifuged then the pallet contains the insoluble part and supernatant contains the insoluble materials like cell membrane and associated biomolecules. In cells the cell membrane is made of phospholipids and if your desired product is associated with cell membrane then pallets resuspended and lipolysis is done to bring the biomolecules separted from the membrane. So here with lipolysis one wants to extract biomolecules attached with the membrane.
