this is a two step process-1) plasmid preparation and 2) gene preparation

1) plasmid preparation- digest the plasmid with suitable RE enzymes so it will linearize.

2) gene preparation - first make cDNA of the gene. amplify the gene using primers which have same RE sites used to cut plasmid DNA.amplify the gene and perform RE digestion.

now perform ligation reaction of plasmid and gene. you got a recombinant plasmid. transform this recombinant plasmid into competent cells. screen the positive transformant and do the further experiments.
