How are you able to ensure that the E. coli you grew for an RNAi experiment contains the plasmid you ordered from the RNAi library?

Group of answer choices

Grow in presence of antibiotic

All bacteria will have it

Grow in presence of GFP

Use PAM sequence

Add IPTG to the culture

Homework Answers

Answer #1

Answers are "ALL BACTERIA WILL HAVE IT" as E.coli is known to have 2 plamsids. The E.coli grown from the rnai experiment are known for resistance to amphicillin thus "GROW IN PRESENCE OF ANTIBIOTIC".

Why not Grow in presence of GFP - green fluorescence protein, it is found in marine organisms mainly, thus it is not a choice fr determining, Use PAM sequence, it is used for the cut of Cripsr locus and the add IPTG that is used to express protein below 16°centigrade.

The genome of the bacteriophage M13 was modified to include a CRISPR-Cas 9 operon with the following features: The operon includes a gene coding for protein Cas 9, and a gene coding a sgRNA. The CRISPR-Ca9 operon is under the control of a bacterial constitutive promoter. The sgRNA gene includes a spacer flanked by two repeats. The sequence of the spacer matches a sequence of an antibiotic resistance gene present in an antibiotic resistant form of a bacteria. Bacteria containing...
