Mechanism:
Viral infection leads to the secretion of TNF-alpha. TNF-alpha binds to its receptor TNFR1. The downstream signaling involves the transmission of the signal from TNFR-associated death protein TRADD to RIPK1. RIPK1 recruits RIPK3 forming necrosome. Ripoptosome phosphorylates MLKL and drives its oligomerization. This leads to the insertion of MLKL into the membrane and increases its permeability. Damage-associated molecular patterns are released from the cell triggering the immune response.
