In which orientation will the Cre-loxP system be if one wanna overexpression a gene?
The Cre-lox system is a technology that can be used to induce site - specific recombination events.
Three general outcome based on the location and orientation of the lox P sites -
1. If the lox P site are on the same DNA strand and are in oppsite orientation, recombination results in an Inversion and the region of DNA between the lox P site is reversed.
2. Deletion - if the site face in the same direction, the sequence between lox P sites is excised as a circular piece of DNA, so no gene expression.
3. Translocation - if the sites are on separate DNA molecules, a translocation events is generated at the lox P site.
Hence gene show overexpression - if the lox P site are on the same DNA strand and are in oppsite orientation, recombination results in an inversion
