ATP synthase synthesises ATP using a proton gradient in the cell .This happens in the mitochondria in case of Eucaryotes . While in Procaryotes this happens in the plasma mebrane.
When the holes are poked in the cell membrane of eucaryotic cell ATP synthase will still function since it is located on mitochondrial membrane . Whereas in case of Procaryotes ,if the holes are poked in plasma membrane ,ATP synthase will not function due to leakage of protons outside the cell .
