The process of creating phosphodiester bonds between an insert DNA and vector DNA is called Ligation and the enzyme that catalyzes the formation of these bonds is DNA ligase .

This is a very common and essential procedure in Recombinant DNA technology and molecular cloning. Insert DNA usually inserted into the vector to make copies. DNA ligase forms phosphodiester bonds between 3' hydroxyl end of one DNA fragment with the another fragment's 5'phosphoryl end. This process needs ATP.
