Examine the following restriction recognition sequence.

a). (One point)Your mission is to complete the entire recognition sequence, which is palindromic. 5’-G/T C _ _ _-3’ 3’-_ _ _ _ _ _-5’

b). (3 points) The dashed line indicates the endonuclease cut site. Draw out the result after digestion with the enzyme, labeling all 5’ and 3’ ends. T

his enzyme leaves (CIRCLE the correct answer): 3’ overhangs or 5’-overhangs

Answer #1

a. Palindromic sequence is the sequence which reads same on both the strands.

Eg: 5' - GAATTC - 3'

3' - CTTAAG - 5'

For the given incomplete sequence:

5' - GTCGAC - 3'

3' - CAGCTG - 5'

b. The restriction enzyme cuts this site by cutting in between G and T from the 5' end. Hence after restriction, the cut strands overhangs will be as follows:

5' - G -3'    5' - TCGAC - 3'

3' - CAGCT - 5'    3'-G - 5'

Hence it produces both the 3' and 5' overhangs.

The 5' overhang is TCGAC and 3' overhang is G.
