The collagen protein consist of mainly glycine, proline and hydroxyproline. The newly synthesized collagen contain only proline and hydroxylation of proline occurs post-translationally with the help of enzyme prolyl hydroxylase. If hydroxyproline is administerd, it is degraded and thus does not added to the collagen molecules. The precursor for collagen molecule is proline only not hydroxyproline. Thus, administerd hydroxyproline is not utilized for collage synthesis rather it degraded and therefore the newly synthesized collagem molecules are not 14C labelled.
