You are required to make a 10ml PBS (Phosphate-buffered saline) sodium phosphate buffer. 7.5pH, 200mM

how many grams of acid and how many grams of base do we need? how much demi water is required?

Answer #1

Sodium Phosphate Buffer is a mixing of Sodium Dihydrogen phosphate NaH2PO4H2O ( Acid) and Disodium Hydrogen Phosphate Na2HPO4-7H2O (Base).

we have to prepare 10 ml of Sodium Phosphate Buffer with 7.5 pH.

  • Add 8 ml distilled water in a container.
  • Add 0.202 g of Na2HPO4-7H2O to the solution.
  • Add 0.034 g of NaH2PO4H2O to the solution.
  • Adjust the solution to get final pH by using Hcl or NaoH.
  • Add distilled water upto volume is 0.01 L.

And finally we obtain 10 ml of Sodium Phosphate Buffer solution.
