5-FU transported in to the cell by a saturable carrier (receptor) based transport system present in the cell membrane.

5-FU binds to thymidylate synthase (TS) enzyme and Interrupt synthesis thymidine, which is a nucleoside required for DNA replication. Thymidylate synthase methylates deoxyuridine monophosphate (dUMP) to form thymidine monophosphate (dTMP).

After binding with substrate dUMP TS enzyme takes on its active form and undergoes a change in conformation.
