mTOR Complex is involved in cell growth and proliferation.mTORC is translocated into the lysosome through Rag GTPase when induced by amino acids.Growth factors also activates phospholipase D (PLD) and phosphatidic acid (PA) for the activation of mTOR signalling pathway.Studies suggests that in the absence of PLD there is absence of translocation of mTOR complex into the lysosome.Activation of mTOR is done in two steps where firstly amino acids translocates mTOR from cytoplasm to lysosome and then GTPase Rheb dependent activation of mTOR is done in lysosome.PLD activates mTOR in response to growth factor and amino acids.On binding with Rheb PLD1 gets activated and is involved in converting phopshotidylcholine to choline and PA.
