Answer #1

Process -

1. Bacteria was grown indefinitely in medium containing heavy isotope of nitrogen, N15.

2. Bacteria was transferred to a medium containing light isotope of nitrogen, N14.

3. After 1st generation all the DNA was parental, that is, containing heavy isotope of nitrogen.

4. After 2nd generation all hybrid DNA was recovered, that is, containing both the isotopes of nitrogen.

5. After 3rd Generation both hybrid and light DNA was recovered.

This lead to the proposal that DNA conservation was a semi conservative process.

This question is about the Mode of DNA replication: Meselson-Stahl experiment 4. The investigators "anticipated that a label which imparts to the DNA molecule an increased density" (Introduction, paragraph 1) might help reveal how atoms in parental DNA are distributed to progeny DNA molecules. What label did the investigators use to be able to detect differences among molecules?

Q3: The diagram below shows the results from the famous Meselson-Stahl experiment proving the semiconservative nature of DNA replication.  Note: after one generation (b) all the DNA was hybrid, after two generations (c), 50% of DNA is hybrid and 50% is light. What would you expect to obtain if the bacteria were allowed to continue to divide:             (4 pts) Results expected after 3rd Generation: Percent Light DNA = __%,    Percent hybrid DNA __% Results expected after 4th Generation: Percent Light DNA = __%,    Percent...

To determine the semi-conservative nature of DNA replication, Meselson and Stahl performed the following experiment: Cells were grown in 15N for several generations and then shifted to 14N medium. Samples of cells were taken before the shift to 14N (generation 0) and then after multiple rounds of replication in 14N. The DNA was isolated from cells and subjected to centrifugation in gradient. The following results were obtained after 2 rounds of replication. Which of the following would result from a...
