In DNA isolation from plant leaves, why was there a need for different concentration of CTAB (cetyltrimethyl ammonium bromide) for the buffer, extraction and precipitation solutions?
CTAB Is a cationic surfactant which helps in the extraction of highly purified DNA from the plant sources. since the plant cell contains cell wall , cell membrane made of lipids and polyphenols .therefore to separate the genomic DNA specifically from the polysaccharides and the lipids a higher concentration of the CTAB is being used in the buffer and the extraction solutions. polyphenols are also to be separated which precipitate along with the DNA molecule hence a comparatively lesser concentrated solution of CTAB is used in the precipitating buffer solutions.
