Why was the slide immersed in acid-alcohol after hematoxylin staining and washing in the H&E procedure? If the acid-alcohol step were omitted, how would the slide look different once H&E staining was completed?

Ans . The slide is immersed in acid-alcohol after hematoxylin staining and washing in the H&E procedure , to remove the excess stain from the slide.

On most occasions, when hematoxylin is employed, a differentiation (destaining) step is required to remove non-specific background staining and to improve contrast.A weak acid- alcohol is used. After this treatment , blueing and thorough rinsing is again required.

If the acid-alcohol step were ommited then non-specific background stains are observed on slides and traces of water are also observed on the slides.
