Describe in details:
molecular details of Ca2+-dependent vesicle fusion with membrane ?
Vesicle fusion with membrane requires impotant proteins such as SNARES,synaptotagmins, complexins, MUNC18, MUNC13, complexins, synaptobrevins. The synaptic proteins synaptobrevin (also referred to as VAMP), syntaxin 1 and SNAP-25 belong to the SNARE protein family. During priming, recruitment of MUNC18,MUNC13 with SNARE protein (syntaxin 1 and SNAP 25 ) occurs. this result in activation of SNARE acceptor complex. The vesicle with synaptotagmin and synaptobrevin tails is attracted towards the SNARE pritein complex. Complexin , another protein arrives to the site and attaches to the SNARE complex along with synaptobrevin of the vesicle. at this time ca2+ influx occurs from inside membrane which triggers the release of MUNC18 and complexin from the complex. Later the existing SNARE complex fuse the vesicle into the plasma membrane.
