Whats the challenge of designing a vaccine for sleeping sickness (Trypanosoma brucei)?

Answer #1

Question:

Answer: African sleeping sickness is caused by Trypanosoma brucei. Trypanosoma brucei is belong to Trypanosoma phylum. But development of vaccine against it quite challenging because of following reasons:

· Antigenic variation: It has millions of variant surface glycoprotein (VSG) in coat. But rearrangement of genes will change the marked VSG due to antigenic variation.

· IgM isotypes: In response to VSG based vaccine, immune system produce IgM antibodies which has very short life time.

· Non-variable surface molecule: Flagellar pocket (FP) based vaccines provide temporary immunity at low infection.

· Suppression of B-cell homeostasis and memory cells: Anti-trypanosoma vaccines leads to suppress the B-cells and its memory cells.

What specific challenge does  Trypanosoma brucei present to the immune system? It frequently changes its antigens, staying one step ahead of the host's antibody response. It specifically attacks B lymphocytes preventing activation and antibody production. It specifically attacks T lymphocytes preventing their activation and crippling the cell-mediated immune response. Its antigens are not recognized by B and T lymphocytes so these host defense cells don't respond to the infection.

You have a research laboratory and are interested in designing a vaccine to prevent infection by a newly identified species of Streptococcus. This is an extracellular bacterium that has been determined to produce a capsule which may be important in infection by preventing phagocytosis. The capsule is not antigenic and so a vaccine composed of just capsule may not elicit a sufficient immune response. Outline what type of vaccine strategy may be employed and justify your answer based on your...
