You have a stock solution of Gel Running Buffer made at 10x of the working buffer. How would you dilute it to 2L of a 1x solution?
We have to dilute 10X buffer to a 2L of 1X solution.
10X means 10 times concentrated.
We have to make it 1X that means we have to dilute it 10 times.
We know that
M1V1 = M2V2
M1 = Stock concentration (10X)
V1 = Volume needed of M1 concentrated stock
M2 = Desired concentration (1X)
V2 = Desired volume (2L)
As per as M1V1 = M2V2
10X * V1 = 1X * 2L
V1 = 0.2 L (200mL)
This means we need to add 2 - 0.2 L = 1.8 L (1800 mL) water in 0.2 L (200mL) of 10X concentrate to make it 2L of 1X solution.
Thus if we add 1800 mL of water in 200mL of 10X buffer we can dilute it to 2L (2000 mL) of a 1X solution.
