The enzyme ABC exists in two different forms, one form is present only in the mitochondria and the other form can only be found in the cytosol. You are asked to purify the cytosolic ABC. Explain how centrifugation might be involved in the process.

Density gradient centrifugation can be used to purify the cytosolic ABC enzyme.

Sequencial centrifugation can be done to fraction out some cell part in each centrifugation. The order of pallet formation in sequencial centrifugation will be : nuclei sediments, dense cell organelles like mitochondria; lysosome: peroxisomes sediments, plama membrane; ER and at last the pallet of ribosomes.

In the supernatant of last centrifugation, there will be presence of cytosolic proteins which will contain ABC enzyme.
