a standard plate count have 25-250 colony forming units. why is it considered as a limitation of the standard plate count?

Answer #1

The reason why 25-250 CF( colony forming units) is considered as a limitation because, some kind of bacteria which are put under examination will have an influence on the size of the colonies and also on the number of them that can be developed on the plate.

The supply of food may also be an issue because colonies close to each other may merge and also there is a chance in which the neighbor colonies may also stimulate their growth.

There are these 2 scientists Breed and Dotterrer who did chose their plates which are countable from triplicate plates from each dilution, but the requiring acceptable plates within 20% on average. Even though the CFU count >300 is totally unsatisfied , but the CFU count <400 is good and satisfiable.

In microbiology, colony-forming units (CFUs) are used to measure the number of microorganisms present in a sample. To determine the number of CFUs, the sample is prepared, spread uniformly on an agar plate, and then incubated at some suitable temperature. Suppose that the number of CFUs that appear after incubation follows a Poisson distribution with μ = 15. (a) If the area of the agar plate is 75 square centimeters (cm2), what is the probability of observing fewer than 4...

Total Heterotrophic Plate Count: A serial dilution was conducted using standard methods, then selected dilutions were used to set up pour plates with total plate count agar, and plates incubated for 48 – 72 hours. Each dilution was plated in duplicate, with 1.0 ml of inoculum placed into Petri plates, then pour plates prepared. After incubation, you counted the following: 10*-3 dilution: Too many to count on both plates. 10*-4 dilution: 378 colonies; 427 colonies. 10*-5 dilution: 53 colonies; 47...

1) In most laboratory applications, for plate counts to be considered valid, they must have between 30 and 300 colony forming units on them. To achieve this, scientists must dilute the sample prior to plating it. If a culture of bacteria is experiencing exponential growth has 8,449 cells per mL, and an instantaneous growth rate constant of 0.5 hours-1, after 2 hours, what is the total dilution that needs to be performed to yield 71 colony forming units on an...

In each case below, you have plated 0.01 mL of the blood or diluted blood on a plate containing a confluent monolayer of tissue culture cells that are susceptible to the virus. You allowed the plates to incubate overnight, and upon arriving at the lab early...
