EMS is an alkylating agent which converts Guanine to O-6-ethylguanine. DNA polymerase catalayse the replication elongation process by adding nucleotide bases. During elongation polymerase enzyme add Thymine into 0-6-ethyl gunanine intsead of Cytosine.

(Note: According to the law Gunanine always pair with cytocine and Thymine always pair with adenine.)

Thus mismatch occur in the sequence and this can not be repaired. If the damage is not able to repair then the cell undergoes cell cycle arrests and followed by apoptosis.
