Membrane filtration is the method for the testing of coliforms in waterways . Samples to be tested are made to go through a membrane filter of particular pore size (generally 0.45 micron). filter surface stops the microorganisms present in water . the filter is put in a petri dish which is sterile and saturated in appropriate medium, growth of the desired organisms is seen, while that of other organisms is suppressed. separate colony are developed by each cell , which can be counted directly, and the results calculated as microbial density. Sample volumes of 1 ml and 10 ml is used for the water testing, for achieving a final desirable colony density range of 20-60 colonies/filter. Contaminated sources require dilution to achieve a countablebmembrane.
