3. Amplify the gene of interest using gene specifuc primers in polymerase chain reaction which is a technique to increase the quantity of DNA

4. Digest a plasmid and gene of interest using same restriction enzyme

5. Ligate the plasmid and gene of interest

6. Transfer the plasmid into recipient strain using heat shock method

7. Once the plamsid goes inside homologous sequences from donar strain chromosomal DNA and plasmid DNA interact to insert the mutant gene instead of normal gene.
