Construct a recombinant bacteria strain with a gene mutated. You need to design and suggest the steps.
1) cloning of gene
2) mutation (how to do it)
3) which homologous recombinant method you use
4) how to verify the strain

Explain the steps in detail.

Homework Answers

Answer #1

1. Treat the donar strain with mutagen

2. Isolate DNA from Donor strain

3. Amplify the gene of interest using gene specifuc primers in polymerase chain reaction which is a technique to increase the quantity of DNA

4. Digest a plasmid and gene of interest using same restriction enzyme

5. Ligate the plasmid and gene of interest

6. Transfer the plasmid into recipient strain using heat shock method

7. Once the plamsid goes inside homologous sequences from donar strain chromosomal DNA and plasmid DNA interact to insert the mutant gene instead of normal gene.

Imagine you have discovered a specific mutation (an A in the gene has been mutated to a T) in several patients that causes them to have neon green tongues. You want to study this specific mutation in mice and plan to use the CRISPR/Cas9 system to allow you to do so. How might you go about doing this?

You are screening newborns to see if they have the progerin gene mutation or not.  Describe how you would use FISH as a diagnostic toolfor this disease. What technique could you use to see if the progerin protein or normal Lamin A protein was being expressed.  For both, explain the technique and how you would determine which protein the patient expressed (the normal or progerin). Everything you need to know about the progerin gene and protein is in the Human Connections section....

Lactose tolerance (the ability to process milk during adulthood) was actually a mutation that arose randomly during evolutionary history. Now, it is estimated that 1/3 of the world's population is lactose tolerant. Explain how this mutant allele might have increased in frequency to reach 1/3 of the world's population. Dwarfism can be caused by a shortage of human growth hormone. Bacteria can be used to produce the protein. Describe the steps you would take to use molecular cloning to insert...

If you are to design a disinfection reactor by adding free chlorine to kill the bacteria and virus (the reaction is typically first-order exponential decay of microbe number with time), do you want to use A. PFR or B. CMFR? Circle your choice and explain your answer in 1-2 sentences

In MultiSim, design a PWM circuit. You will need a 0 to 9 counter, which you should build from flip flops and basic logic gates. The PWM input will come from 4 input switches as a binary number. For the comparator, you can build your own from basic gates, or use one of the commercially available comparator chips (74HC85 Digital IC). An oscilloscope in MultiSim can be used to verify duty cycles.

You are studying disease X, a genetic disorder that arises from a trinucleotide repeat expansion mutation which is a mutation where entire codons are multiplied during the replication process. The mutation is found in the CaM gene, that is a 450-base pair (bp) sequence. The trinucleotide repeat expansion results in the addition of 10 codons (30 bp) at nucleotide 150 (ie in the centre of the gene, away from the 5’ and 3’ end). The sequence of the normal gene...

Part 1: Click on the box that says “Expression.” You are presented with a gene that contains a regulatory region and a transcribed region. Your goal is to produce 3 proteins from each of three genes. The proteins are indicated by the shapes in the box at the top right. You need to successfully transcribe each gene, and then translate the message from the gene into a protein using your knowledge of how these processes work. GENE 1 Questions What...

1. Create a voltage divider. Design a circuit which can provide a fraction of the voltage ( x V) out of the regular batteries. The equipment you can use: 2 D-batteries (about 1.5V each) and one piece of nichrome wire and some connecting wires. If you need to make 2V, how would you connect it? Draw a detailed circuit to show the connection. Explain in detail why it works. Use a voltmeter to check your design. Draw the voltmeter on...
