simultaneously IP3 opens calcium channels and increases cytoplasmic calcium level which combine with protein kinase C and initiate biological response while DAG (diacetyl glycerol )  act as secondary messenger that also activates enzyme called Phosphokinase C  that phosphorylate a bunch of enzymes and cause muscle contraction....Thanks
