Explain what would happen druing a PCR elongation step if a dideoxyribonucleoyide get incorporated into a growing nucleotide chain. why would this happen?

Homework Answers

Answer #1

A deoxyribonucleotide (dNTP) does not contain a hydroxyl group at the 2nd carbon atom.
A dideoxyribonucleotide (ddNTP) does not contain hydroxyl groups at both the 2nd and 3rd carbon atoms.

The elongation step of the PCR reaction involves the formation of phosphodiester bonds.
A phosphodiester bond is formed between the 3'-OH group of one nucleotide and the 5'-phosphate group of the other nucleotide. Since a ddNTP does not contain a 3'-OH group, the chain can no longer be extended once it is incorporated into the chain.
