pH of the buffer is defined as the pKa or the negative log of the dissociation constant of the acid.All buffers have an optimal pH.. A buffer consists of a weak base and its conjugate acid or weak acid and its conjugate acid.Optimal pH is the pH of an enzyme at which it is most active . pH =pKa +log 10 (A-)/(HA) .This is Henderson hasselbalch equation by which the pKa can be determines.

When added to the solution , buffers are molecules that hav ethe ability to donate or accept protons to resist changes in pH as acids or bases .
