For the histology purpose we use Haematoxylin and eosin stain, also known as h and e stain,

The stratified squamous epithelium is present over the skin surface and this stratified nature is visible because of presence of keratin at the the topmost layer the white space is is seen because of this covering of the keratin protein over the stratified squamous epithelial cells of the skin the white space is also appear because of the adipocytes/ fatt cells,because fat cells are leopard in nature so this sadness do not update the the h and e stain and appearance white space or empty space in the histology slides.
