Why would different primer sets require different PCR conditions?
In PCR (polymerage chain reaction) . For the replication of DNA (formation of new DNA copies from parent DNA strands Requirement for PCR is DNA polymerage enzyme :this will work at some requirements :1)It will be work on single strand of DNA. 2)At the starting point their should be double stranded . To complete 2nd requirement their is RNA primer is needed . Primer is formed for RNA polymerage. This RNA primer is read the DNA strand in 3'-5'direction , polymerage RNA primer in 5'-3'direction . So, their is multiple primers are formed to initiate replication process . On leading strand (3'-5')only one primer is formed and their is continuously DNA polymerisation takes place. But, lagging strand(5'-3')their we need multiple primers because replication fork is on 5'end So, lagging strand is formed in multiple fragments .
