Ansa) :-

A midstream specimen of urine (MSU) is best, as the first bit of urine that you pass may be contaminated with bacteria from the skin. And it also contain epitheloid cells of urethra lying which can give false results.

Ansb):- each strip contains various reagent which work by adsorption of urine in the strip paper by process called chromatography.

So if lead is left open it will damage the reagents and water may get adsorbed in the strip paper making it ineffective.

So avoid exposure of strips to moisture, volatile fumes, or direct sunlight.
