The contraction of the rat ileum longitudinal muscles mediated by the 5-hydroxytryptamine receptor. 5-HT and alpha-methyl-5HT equipotently induced contractions, whereas 5-methoxytryptamine and 2-methyl-5-HT were less potent.

Neither tetrodotoxin nor atropine affected the 5-HT contraction, indicating a smooth position of these 5-HT2 receptors in the muscles.

The involvement of either a selective antagonist 5-HT2B (SB 204741), 5-HT3 (granisetron) or 5-HT4 (SB 204070) influenced the contractions to 5-HT slightly. Thus, in all subsequent experiments, they were also included in the organ bath solution to isolate the principal contractile component pharmacologically.
