Staining is a technique used to enhance the contrast of the microscopic image.
Staining can be of two types : Simple staining and differential staining.
Of these, gram staining comes under differential staining and it is used most frequently to differentiate bacteria into gram positive and gram negative.Step by step procedure is explained below.
*Apply primary stain to heat fixed bacterial cultured slide. The primary stain used is Crystal violet
* Add iodine, which fixes the crystal violet to cells.
* Decolorize with ethanol or acetone.
* Counterstaining with Carbol fuchsin.
Result: Gram positive organisms retain purple color where's gram positive will have pink color. ✅
